Three new species of Agaricales from eastern Honshu, Japan, are described and illustrated. (1) Clitocybe minutella sp. nov. (section Vernae), having white, very small, clitocyboid basidiomata with white, strigose mycelial tomentum at the base of the stipe and trichodermial
elements in the pileipellis, occurs on leaf litter in deciduous oak forests. (2) Crepidotus longicomatus sp.nov. (section Echinospori), forms very small (up to 3.5 mm in diameter), reniform basidiomata densely covered overall with white to pale yellow, erect,
thick-walled, long hairs. It has melleous, echinulate basidiospores, and was found on fallen dead branches in lowland oak
forests. (3) Crepidotus virgineus sp. nov. (section Crepidotus), forming pure white, reniform basidiomata, has smooth, ellipsoid basidiospores, subfusiform-pedicellate to irregularly cylindrical
cheilocystidia, and abundant clamp connections. It occurs on fallen dead branches in lowland oak forests.

Background
Lectins are a diverse group of carbohydrate-binding proteins exhibiting numerous biological activities and functions.Methods
Two-step serial carbohydrate affinity chromatography was used to isolate a lectin from the edible mushroom clouded agaric (Clitocybe nebularis). It was characterized biochemically, its gene and cDNA cloned and the deduced amino acid sequence analyzed. Its activity was tested by hemagglutination assay and carbohydrate-binding specificity determined by glycan microarray analysis. Its effect on proliferation of several human cell lines was determined by MTS assay.Results
A homodimeric lectin with 15.9-kDa subunits agglutinates human group A, followed by B, O, and bovine erythrocytes. Hemagglutination was inhibited by glycoprotein asialofetuin and lactose. Glycan microarray analysis revealed that the lectin recognizes human blood group A determinant GalNAcα1–3(Fucα1–2)Galβ-containing carbohydrates, and GalNAcβ1–4GlcNAc (N,N'-diacetyllactosediamine). The lectin exerts antiproliferative activity specific to human leukemic T cells.Conclusions
The protein belongs to the ricin B-like lectin superfamily, and has been designated as C. nebularis lectin (CNL). Its antiproliferative effect appears to be elicited by binding to carbohydrate receptors on human leukemic T cells.General significance
CNL is one of the few mushroom ricin B-like lectins that have been identified and the only one so far shown to possess immunomodulatory properties. 相似文献4.

Armillaria root rot, caused by Armillaria tabescens and Armillaria mellea, is a major cause of premature tree death in peach orchards in the southeastern United States. The root systems of infected trees can become entirely colonized by Armillaria, serving as an inoculum source for adjacent trees and providing massive inoculum levels in replant situations. If dead or dying trees could be colonized by an effective competitor of Armillaria before their removal, the extent of root colonization by the pathogen could be reduced, thus decreasing the threat to adjacent trees and/or subsequent plantings. Interactions between five species of saprobic lignicolous fungi (Ganoderma lucidum, Hypholoma fasciculare, Phanerochaete velutina, Schizophyllum commune, and Xylaria hypoxylon) and the two Armillaria species were examined in controlled conditions to provide proof of concept for competitive exclusion of Armillaria from peach roots. On agar-coated glass slides, all five potential antagonists induced detrimental reactions in >58% of the Armillaria hyphae observed, with the majority resulting in hyphal swelling or granulation. On poplar wood blocks, all antagonists consistently either overgrew Armillaria colonies or—in the case of S. commune—engaged in deadlock reactions; in all cases, the viability of Armillaria colonies was reduced to <30% of that of unchallenged controls. When inoculated simultaneously onto opposite ends of peach root segments, all antagonists consistently reduced growth and viability of Armillaria on and under the bark, whereby reduction of pathogen growth underneath the bark, Armillaria’s primary ecological niche, was most pronounced for G. lucidum, S. commune, and X. hypoxylon. When root segments were allowed to be colonized entirely by Armillaria before being inoculated with the antagonists, the latter were able to overgrow the pathogen on the root surface but unable to pre-empt it from underneath the bark. In summary,G. lucidum, S. commune, and X. hypoxylon caused strong hyphal and mycelial interference reactions and the most pronounced reductions in growth of Armillaria above and below the bark, indicating that they would be the most promising candidates for field-scale evaluations to restrict colonization of dead or dying peach trees by Armillaria in the orchard. 相似文献

Clitocypin and macrocypin are cysteine protease inhibitors of the mycocypin family which is unique to basidiomycetes. We have established that Clitocybe nebularis and Macrolepiota procera each contain genes for both macrocypin and clitocypin. Both are expressed in M. procera but only clitocypin in C. nebularis. Further analysis of mycocypin expression at the mRNA and protein levels in mature fruiting bodies of M. procera revealed that clitocypin is expressed evenly throughout the fruiting body, while the level of expression of macrocypins varies, and, at the protein level, is much higher in the veil fragments and the ring. The expression patterns of various mycocypins were determined in Coprinopsis cinerea, using promoters linked to a reporter gene. The expression profile of the clitocypin promoter was similar to that of the constitutive promoter gpdII from Agaricus bisporus, while that of the macrocypin 4 promoter was limited to the outer edges of the fruiting body throughout development. In addition, the activity of the macrocypin 3 promoter was different, indicating different regulation of expression for different macrocypin genes. The complex, tissue specific expression patterns for mycocypin genes suggest different biological roles for the products, either in regulation of endogenous proteases or in defense against pathogens or predators. 相似文献

Recent and still little known fungus poisonings This research paper gives an overview of recent fungus poisonings, their symptoms, and, as far as known, their toxins. Intoxications often are the consequence of the confusion with edible or “medicinal” mushrooms. The toxic species Freckled Dapperling (Lepiota aspera), Angel's Wing (Pleurocybella porrigens), Jack O'Lantern (Omphalaotus olearius) and Clitocybe amoenolens are found in Central Europe. It is little know that a rich morel dish can cause neurologic symptoms. Species from Asia became acquainted, that can provoke most serious intoxications. The “Yunnan Sudden Unexplained Death‐Syndrome” could be explained by the consumption of the “Little white mushroom” (Trogia venenata). The often fatal rhabdomyolysis after the consumption of Russula subnigricans can be traced back to the effect of Cycloprop‐2ene‐carboxylicacid. Particularly fatal is the confusion of the mushroom “Reishi” (Ganoderma lucidum), highly esteemed as the “mushroom of immortality”, with Ganoderma neojaponicum or with Podostroma cornu – damae, the most poisonous mushroom worldwide. 相似文献
